recombinant non-replicating adeno-associated virus vector of synthetic liver-tropic serotype 843 (rAAV843) encoding a codon-optimised truncated (B domain-deleted) variant of human blood coagulation factor VIII (hF8) preceded by a Kozak sequence and with expression under control of a synthetic liver-specific promoter (Lxp-2.1) and a synthetic miniature polyadenylation signal based on the rabbit beta-globin polyadenylation signal; flanked by adeno-associated virus 2 (AAV2) inverted terminal repeats (ITRs). An artificial 8 amino acid linker sequence connects the N-terminal part with the C-terminal part of the F8 transgene
