VCN-01 contains 4 genetic modifications with respect to the wild-type virus (HAd5): i) a modified E1A promoter with E2F and SP1-bindin boxes (promoter 415); ii) the delection E1A-Δ24 (24 base pair deletion in the E1A gene); iii)the replacement of the KKTK domain in the adenovirus fiber protein by the RGDK amino acid sequence; iv) and encodes the soluble form of the human sperm hyaluronidase (PH20) transcription unit
