That is a great question. The short answer is that yes- we need more data. We need more sequence from control individuals without so that we have a better understanding of the extent of variation within the population. Additionally, we need improved models so that we can better understand how non-coding variation impacts phenotypes. Understanding how environmental factors impact disease is also a significant challenge in understanding biological phenotypes.
